This is a man beater killer defense designed to take away option routes, crossers and slants over the middle by giving the defense both a deep eraser and an underneath spy, so your primary read as the defense is watching the quarterback eyes and letting the hole defender trigger on any in cut or drag before the receiver can settle into the void.
Pre-snap, check the safety alignment: if the offense sees single high safety look and shifts protection or checks to a max protect play-action, expect them to test the deep half since Cover 1 Hole only has one deep defender. Watch receiver splits and stack alignments before the snap, tight bunch or stack formations are designed to beat man coverage with rub routes, so be ready to jump switch assignments with your corners. Post-snap, key the quarterback's first read: if his eyes go to the middle of the field early, trust your hole defender to sit on any drag, slant or in-breaking route rather than following the receiver vertically.
